The effect of cleaning cycles and a biocide on the reduction of flux due to fouling was investigated.
We address this problem through a computational approach, which considers the effect of drug application within a generalized biochemical pathway and by studying the effect of changes regarding the type and strength of inhibitors on the reduction of flux.
It is apparent that lack of CA IX causes a marked and statistically significant reduction of flux from about 1.8 mM/min in WT to ∼1.2 mM/min, almost identical to the reduction caused by lack of CA XIV.
While a high sensitivity of lactate dehydrogenase towards NADH/NAD + [ 27] was not observed here, the reduction of flux towards acetyl-CoA (J3) by inhibition of pyruvate dehydrogenase, and therefore also the ethanol flux (J6), in response to the increase in NADH/NAD + was indeed observed [ 27– 30].

Despite a reduction of the flux into the oxidative branch of the PP pathway, the flux through PHK remained largely constant (29.2 vs. 32.8 when phk was over-expressed).
Next we tested if individual simulations show overflow-like metabolism or not. Figure 4 shows a frequency diagram of the reduction of the flux through the high-yield pathway at maximal growth rate, as a percentage of the maximal flux through the high-yield pathway.
Quantitative analysis of the autophagic flux, expressed as the ratio of LC3-II signals in presence and absence of BafA1, showed a significant reduction of autophagic flux in the four sarcoma cell lines exposed to Tv6, although this was not statistically significant for RH cells.
