Reader conditions were as follows: 481 nm excitation, 509 nm emission, gain of 50, and read height of 7.0 mm.
The actual focus height had only minimal effect on the detection of spot numbers but we found that the lower the read height, the greater the fluorescence measurement.
Regions with sufficient read coverage were identified using H, L and G parameters, which are read height, length in base pairs and gap permitted in the length parameter, respectively.
