The obtained iron-starvation dependent phleomycin resistance indicates the potential use of this promoter for selection marker recycling.
This was ligated to a strong splice-acceptor lacZ expression cassette (designed to splice onto exon three of Mro and act as a reporter), and a neomycin resistance gene under the control of the PGK promoter for selection purposes.
Our findings also suggest that care should be taken in using a weak promoter for selection if a simple insertion pattern is desired.

The weaker ARG4 promoter used for selection marker cassettes enables selection at lower concentrations of Zeocin™ (i.e., 25 instead of 100 μg/ml) without obtaining false-positive clones.
The expression vectors included a Neo resistance gene driven by the SV40 promoter for clone selection.
