Responses were assessed every two cycles until disease progression, or more frequently if early progression was suspected.
In patients with castrate-resistant disease, both BAP and uNTx progression occurred more frequently than bone scan progression (∼25% vs ∼25% vs ∼11%, respectively).
However, there were no significant differences between the observed and expected contributions of the thoracic segment to progression (16% vs. 14%, P = 0.70), whilst progression was observed more frequently than expected in the cervical spine (55% vs. 43%, P = 0.09), and less frequently in the lumbar spine (29% vs. 43%, P = 0.04) (Table 6).
We advocate the creation of progression endpoints that utilize all available progression data rather than early fixed timepoint analyses and show that little is to be gained by assessing progression status any more frequently than would be required in routine clinical practice.
Radiological progression was observed more frequently in case of persistent synovitis after 4 months (15.7% vs 7.5% for clinical evaluation (OR=1.70 (0.93–3.12), p=0.086); 19.3% versus 6.5% for US-GS evaluation (OR=3.14 (1.50–6.55), p=0.002) and 21.1% vs 8.0% for US-PD evaluation (OR=2.79 (1.19–6.56), p=0.019) in joints with, versus without persistent synovitis, respectively.
