The enzymatic lysis buffer was prepared composing of 20 mM Tris.Cl, pH 8, 2 mM sodium EDTA, 1.2%Triton® X-100; and immediately before use, add lysozyme to 20 Mg/ml.
Approximately 1 mm square tissue pieces were cut and placed in 35 mm petri dishes containing 1.2 ml freshly prepared Luciferase Culture Medium composed of 1.2 ml of Dulbecco's Modified Eagles Medium (Sigma) with 10 mM HEPES pH 7.2, 2% B27 (Gibco), 25 units penicillin, 25 µg streptomycin and 0.1 mM beetle luciferin (Molecular Imaging Products) [48].
