The ρ status was verified by measuring mtDNA content and four mitochondrial functional markers: mitochondrial membrane potential, oxygen consumption rate, cytochrome c oxidase (COX) activity, and intracellular superoxide content [see Supplemental Material, pp. 3 4 (http://dx.doi.org/10.1289/ehp.1104287)].
In addition, the expression of the transgene was also associated with excessive accumulation of ROS, a declined cellular ATP/ADP ratio, a reduced mitochondria membrane potential and a lower oxygen consumption rate.
