For flow cytometric staining, 100 µl of venous blood was incubated for 20 minutes at room temperature with the optimised amounts of the following fluorescent-conjugated antibodies: anti-αβ-TCR-FITC (clone WT-31), anti-Vδ2-PE (clone B6), anti-CD3-PerCP (clone SK7), anti-CD8-PE-Cy7 (clone RPA-T8), anti-γδ-TCR-APC (clone B1), and anti-CD4-APC-Cy7 (clone RPA-T4).
The PCR master mix (Quantitect, purchased from Qiagen, 2004b) contained optimised amounts of SYBR® Green dye to which primers (5 pmol tube−1) and DNA template (5 μl) were added.
In order to acquire fully quantitative, high signal-to-noise 13C NMR spectra of ethylene-1-octene copolymers in a relatively short period of time, a detailed protocol has been developed, based on the addition of an optimised amount of the relaxation agent chromium III triacetylacetonate and without using nuclear overhauser enhancement (NOE).
Thereafter, 70 g/L of synthesised MCT β-CD was applied on cotton at five pH levels (4, 6, 8, 10 and 12) through padding with 80%% expression followed by air drying, curing at 150 °C for 5 min, rinsing in cold water to remove any superficial MCT-β-CD, spraying of oils in optimised amounts and subsequent drying of treated cotton.
Six factors with a large impact on the MIP synthesis and its analytical performance have been optimised: the amount of template, the type and the percentage of functional and cross-linking monomers, the polymerisation method (i.e. thermal or UV initiation) and the porogenic solvent.
Those methods were different from the ones evaluated in this report, and were not optimised for picogram amounts of input RNA.
In the top-view image, the dark areas are pores that have an average diameter of approximately 20 nm. Figure 2a,b,c,d,e,f shows the morphology of CNTs grown on the optimised NPSiT using different amounts of catalyst.
One of the most serious bottlenecks in developing natural products from marine sources during the past decades has been the availability of biomass and/or of optimised cultivation conditions to gain sufficient amounts of compound for preclinical and clinical studies (Raghukumar 2008; Pan et al. 2008; Schulz et al. 2008).
However, in phage display, we see the major advantage of the scFab format in the context of an optimised procedure for the generation of large amounts of binders on a proteome wide scale [ 30].
It needs to be optimised to achieve reproducible high labelling efficiencies (LE) and specific activities (SA) of recombinant proteins, without wasting large amounts of protein and 99mTc.
These results show that greater amounts of pDNA can be obtained more economically with minimal extra effort simply by using a medium optimised for pDNA production.
