Remarkably, we observed this sequence also at the insertion sites of the distantly related IS identified in the uncultured Desulfobacterium sp. (IS Desp5).
Harbinson et al.[ 25] identified a Thi2p binding domain by genome-wide chromatin immuno-precipitation and we observed this sequence in the promoter region of both TNP1 and AAD14.
Nappe, who co-authored the report with Christopher Hoyte, explained that the doctors simply observed this unusual sequence of events, documented it and alerted the medical community that it is worth studying a possible relationship between cannabis and the child's cause of death, myocarditis, or inflammation of the heart muscle.
But we did not observe this homologous sequence using the all three methods: the genomic Southern analysis, the BLAST analysis of the sequenced genomes of maize, and the PCR amplification, in contrast to an expectation that can be made given the highly constraint detected from this gene.
For example, if an alternatively spliced isoform were generated p = 10% of the time, n = 10 transcript sequences would give a 65% probability of observing this event, and 22 transcript sequences would be required to have >90% probability of observing the event.
To identify genes that had their expression or transcript structure deregulated by insertions (and the mutation clonality required to observe this) we sequenced RNA samples from a subpanel of 26 BCL2 transgenic tumors.
Observing this many distinct LWS opsin sequences in a single individual guppy is a highly surprising result.
Importantly, BM2 translation followed the same pattern in mRNAs where the UAAUG motif was moved as a unit, with ablation of synthesis observed when this sequence was moved any further than 24 nt downstream of its original site (Figure 5B).
Next, using the DNASTAR Lasergene MegAlign program, we performed protein sequence alignments and observed that this sequence is fully conserved in the GnRH-RI protein expressed in the chimpanzee while in the mouse and rat, five of the seven residues in the primate sequence are found at the equivalent site (Table 1).
However, a weak similarity to canonical CENP-B box has been observed within this sequence for four periodically spaced 28-bp repeats (data not shown).
