Confocal laser microscopy coupled with fluorescence- and Raman spectroscopy was used to investigate optical properties of single silver nuclei (clusters).
Nuclei clusters, mitotic cells, and apoptotic cells were filtered from the total cell population using an intersection of DNA content and DNA clusters gates.
We also extended these studies to differentiated primary mouse muscle cell cultures; we observed two types of myotubes in primary cultures: thin slowly contracting myotubes with nuclei aligned along the middle of the fiber (immature myotubes), and thicker, faster contracting myotubes often with small or large nuclei clusters (mature myotubes).
Interestingly, even in very long-term (two week) cultures, thin, slowly contracting myotubes without nuclei clusters persisted in the culture.
Moreover more myotubes with nuclei clusters and larger nuclei aggregates were observed when myotubes were co-cultured with neurons [33].
In some nuclei, clusters of dots representing many copies of MET gene were apparent.
