Myelinated axons without overt myelin aberrations had normal myelin thickness as assessed by growth ratio (axon diameter/fibre diameter) measurements in 10-week-old animals (Fig. 4C).
This inability to restore normal myelin thickness in remyelination with age was associated with a reduced macrophage/microglial response.
The control image (Fig. 5(d)) shows myelin sheaths with normal myelin thickness, as expected from healthy nerves.
Erbin is mostly expressed in regions where myelination is abundant and erbin null mice have hypomyelinated peripheral nerves, suggesting it is necessary for normal myelin thickness of peripheral nerves [ 20].

Fig4 Floxed/Floxed mice have normal physiology and are indistinguishable from controls since they display normal nerve morphology including myelin thickness, and the absence of cell vacuolization in all cells analysed in both CNS and PNS also in aged mice (data not shown and as previously reported) (12).
Fig4 plt/+ heterozygous mice are normal as no vacuolization has been observed in any tissue analysed and myelin thickness is normal in peripheral nerves, as previously reported (3, 18).
Similarly, the acquisition of normal rates of nerve conduction in older Δ PDZ-Prx nerves appears to be a result of Schwann cells having elongated sufficiently to allow for optimal conduction, together with the normal increase in axon diameter and myelin thickness that occurs during development.
To visualize the structural details of myelin sheaths and quantify the myelin thickness, some myelinated axons were examined at higher magnification (× 21,000).
The g-ratio of a normal mature myelinated axon is constant at ∼0.7 since myelin thickness is tightly regulated according to axon size.
However, 12 weeks after transection, efficient remyelination was observed in conNrg1 mutants (Fig. 6A C), in particular an equivalent proportion of axons were remyelinated (Fig. 6B), and myelin thickness was restored to normal levels (G-ratios mean ± SEM; vehicle control 0.653 ± 0.008 conNrg1 0.619 ± 0.013) (Fig. 6C).
