Twelve different truncating SPRED1 mutations were found in 13 unrelated probands: 7 frameshift and 5 nonsense mutations, 11/13 cases were familial.
62, 63 In most cases, nonsense or frameshift mutations cluster within a hotspot region in the last exon, resulting in truncated NOTCH2 proteins that lack degradation signals in the proline, glutamic acid, serine, threonine-rich domain and thus act in a gain-of-function manner (Fig. 1).
