Bleb formation was shown to be critically dependent on the level of myosin contractility [ 4,5,18,26] and increasing the contractility in cells that normally show rare, or no blebs appears to be sufficient for inducing blebbing [ 18,27,28 ].
Moreover no blebs were observed on the cultured cells submitted to ET 10−7 M for at least 20 min (not illustrated).
Optical sections through the center of the cell revealed no blebs of the membrane (Fig. 3b DEF) in contrast to sections 40 min after mitosis (Fig. 3a DEF).
As seen in Fig 5B, essentially no blebs are observed at 25 min (i.e., 20 min after MTX) and little or no dye uptake is seen out to 40 min. Thus, the major morphological changes associated with MTX challenge are blocked by U73343.

Most importantly, no bleb-related adverse events are associated with this procedure.
Only those recordings from healthy looking cells with distinct borders, uniform striations and no membrane blebs or granularity were included in the analysis.
In the extreme case, fluid cannot leave the bleb and the bleb is no longer an outflow pathway from the eye and the eye IOP is equivalent to that of an unoperated eye.

Samples from later time points, however, showed no evidence of blebs and, as expected, contained more lysed bacteria (data not shown).
