The most striking feature of the usual MRI changes is the globoid appearance of the first part of the oculomotor nerve on emerging from the midbrain, indicating a gross localized enlargement of the nerve, consistent with intraneural edema as reported in some experimental demyelinating neuropathy and in chronic inflammatory demyelinating polyneuropathy (CIDP) [28].
A significant axonal regeneration was observed in rats with a preconditioning lesion of sciatic nerve, consistent with previous studies.
In addition, reduction in MBP staining is observed in p27KIP1 knockout mice in the optic nerve, consistent with the requirement for p27KIP1 and PNS myelination [43].
Cumulative g-ratio plots showed complete separation between control and MLD nerves consistent with evidence of thinned myelin in all patients (Fig. 4, Table 4).
As shown in Figure S3I-L, postsynaptic AChR clusters of wildtype size were aligned with the presynaptic nerve terminals, consistent with the well-documented ability of regenerating axons to restore NMJs after nerve injury in adult muscle [20].
More recently, this structure has been described through anatomical dissection [1, 3, 10, 13, 14, 22], histological analysis (demonstrating the existence of parallel collagen bundles and nerve fibres consistent with a ligamentous structure) [1, 3], radiological studies [23, 24, 25, 26], and in association with the pathognomonic Segond fracture seen in association with ACL injuries [27].
The neurological findings are described in supplementary Table 2. Electrophysiological studies revealed peripheral neuropathy with slowing of nerve conduction consistent with a demyelinating etiology in 10 of 15 subjects.
An electromyogram (EMG) showed normal sensory nerve function, low amplitude on motor stimulation, mild (10%) decremental response on repetitive low frequency, and incremental response on high-frequency nerve stimulation, consistent with botulism (13 ).
In the present study, GluN2B was found to be upregulated in RNA and protein levels after sciatic nerve ligation, consistent with the previous studies in different chronic pain models [ 28– 30], which suggests the possibility of using this mechanism as a target for the NP treatment.
The smaller number of U-turns in the Stat3-ca/Y27632 and CNTF/Y27632 groups, as compared with the CNTF-treated nerves, is consistent with a local attenuation of growth-inhibitory cues by the blockade of ROCK.
The clinical manifestations reported in the studied brothers in the form of tall stature, distorted facies, orodental findings, syndactyly, and encroachment of cranial nerves are consistent with sclerosteosis and mutation in SOST gene.
