Since most D2.50N mutations show lower agonist affinity and become less active in response to agonist (Wilson et al., 2001), a protonated D2.50 appears to be incompatible with agonist binding, thus deprotonation is favored.
The unaffected mother (NIJ-001) who only carried the T22M mutation, showed lower amount of the isoform 1, and normal amount of isoform 2. These results are compatible with lack of FBXO7 protein expression from the allele containing the IVS7+1G/T mutation, and with only isoform 2 being expressed from the allele containing the T22M mutation.
As shown by the susceptibility testing, all four strains with the A2063T mutation showed lower MICs for AZM (0.25 μg/mL), and actually, the eight patients with A2063T mutation who were treated with AZM showed complete resolution of clinical symptoms.
Two of three arp-1 lines, with the substitution mutation in Arp, showed lower, though still detectable, amounts of RT-PCR product, and one showed no detectable product (Fig. 2).
It was striking that for three different drugs, isolates with the A90V and D94A mutations showed a lower level of resistance, whereas isolates carrying the D94G and D94N mutations showed a higher level of resistance.
