No disease-associated mutations of γ-synuclein gene or obvious cases of idiopathic human γ-synucleinopathies have been reported so far and the amino acid similarity between human and mouse proteins is high.
Microscopic analysis of these cells revealed that Sso1p(S23S24S59S79) or Sso2p(T28S31S34) alanine or glutamic acid mutations display no obvious defect in pseudohyphal growth (Figure 4).
Most of these mutations cause obvious defects in the pollen grain and affect early stages of pollen development.
In addition to rare driver mutations of obvious importance, there are numerous passenger mutations found at low allelic frequency within the tumor population, presumably due to ongoing genetic stress within the tumor that results in tumor heterogeneity [ 5, 7, 12, 13].
Though a small percentage of Alzheimer's cases arise from genetic mutations that cause obvious disease before the age of 65, the vast majority of people who develop the condition do so later in life from undefined triggers, some thought to be genetic.
These sequences might differ in single point mutations (as sometimes obvious in MLST, e.g., Taiwan Clone isolates may have different gmk alleles), in the composition and sequence of single loci (such as the variable part of the spa gene or the dru region within the SCCmec element) or in the presence or absence of complete genes or multi-gene mobile elements.
Ser162Ala and Ser474Ala mutations had no obvious effect on 14-3-3 14-3-3 14-3-3K4 under any conditions tested (not shown).
When the mutations were not obvious because of location near the sequencing primers, sequencing from the other direction was done to solve this issue.
For example, even when looking at a dataset with close to 500 samples such as the TCGA ovarian, spatial patterning of mutations is not obvious (see Fig. 1, left panel).
The prevalence of TERT promoter mutations, their intuitively obvious importance for cancer development and progression, and the resulting therapeutic potential, will undoubtedly unleash significant efforts to answer these questions in the near future.
