We have previously shown that the fitness of drug-resistant Mtb depends both on the specific drug-resistance-conferring mutations and strain genetic backgrounds, and that compensatory mutations might mitigate the potential deleterious effects of drug resistance on bacterial fitness [8], [18].

We found that Top2 was at low levels or absent in 7 strains, including all 4 strains carrying nonsense mutations and 3 strains carrying missense mutations, while the remaining 7 strains carrying missense mutations displayed intermediate to high protein levels.
This prediction is borne out both by strains bearing Scc4-conserved patch mutations and by strains lacking CHL4 (Fernius and Marston, 2009).
