Nucleotide sequence analysis revealed a single nucleotide mutation at codon 545 of PIK3CA in MKN1 and AGS cells (E545K and E545A, respectively, Additional file 11c), which suggested that AKT is potentially activated in these cells through mutational activation of PIK3CA.
These issues not withstanding, we feel it is fair to say that these mutations do not meet conventional criteria for mutational activation of receptor kinases.
Our data suggest that mutational activation of PKD1 is not sufficient to activate mTOR and that other events which may be mediated by TSC2 are required to activate mTOR in ADPKD cysts, thereby leading to cyst progression.
