For lung cancer patients, detection of activating mutations in EGFR will identify patients most appropriate for first-line treatment with EGFR TKI therapy (Kobayashi et al, 2005; Lynch et al, 2004; Paez et al, 2004; Pao et al, 2004; Zhu et al, 2008).
Although NGS will not identify biomarkers and drug targets directly, it will identify mutations that will guide the development of biomarkers and drug targets.
TILLING (Targeting Induced Local Lesions in Genomes) is a relatively novel reverse genetics technique based on the use of a mismatch-specific enzyme that will identify mutations in any target gene through heteroduplex analysis [ 22].
