It is therefore possible that, given the strikingly similar domain organization, mammalian ortholog of the cytoplasmic PNGase, besides its PNGase activity, may also exert its functions through the similar deglycosylation-independent mechanism, and indeed it was suggested by our observation that the defect of Pngl mutation was rescued by the expression of Ngly1, a mouse ortholog of Pngl (Table 2).
