By the second time point sampled in WEAU and CH40, and the third time point in SUMA, the viral population diversity had expanded dramatically and variants had become structured into clusters, each comprising a single-escape-mutation majority form surrounded by far less common single and double secondary mutations scattered throughout the region sequenced (Fig. 2 and Table S3).
Even though a considerable number of PRKN mutations are dosage mutations, the majority are sequence variants, hence, we decided to perform the initial screen of our cohort only for these variants.
However, in contrast to site 1 and site 2 mutations, a majority of the mutations introduced into site 3 augmented Wnt signaling compared to wild-type levels, with A96R showing the most pronounced effect.
