However, by forcing the de novo functioning of the OXPHOS system, a severe delay in the biogenesis of the respiratory chain complexes was observed in the mutants, which demonstrated a direct functional effect of the mitochondrial genetic background.
Again the exception was the Δ AfchsG and Δ AfchsC Δ AfchsG mutants, which demonstrated an 80%to90%0% reduction in growth in the presence of caspofungin (Fig. 5C).
These results suggest that mutants which demonstrate diploid specific sensitivity to DOX have defects in recombination and/or checkpoint functions which severely inhibit cell cycle progression in G1 or during G1/S transition.
As expected for the diploid ctf4Δ mutant which demonstrates a chromosome loss phenotype, all single isolate colonies demonstrated a high rate of mating to either the MATa or MATα tester strains suggesting that either copy of chromosome III can be lost due to the high rate of malsegregation previously observed in these mutant cells.
Confirmation of this was obtained by analysis of the sarA/alsSD double mutant UAMS-13000), which demonstrated that concomitant mutation of alsSD and sarA had no further impact on the production of acetoin (data not shown) but did reduce biofilm formation even by comparison to an alsSD mutant (p<0.001) (Fig. 4).
