Intriguingly, we observed that Gas2 mRNA decay in Ski2Δ mutant strains was observed only transiently, peaking at 30 min after ER stress induction.
For three individual mutant strains we observed 36, ∼38 and 32% of all cells in the supernatant (Fig. 4).
A similar absence of differences between the WT and mutant strains was observed when invasion of porcine BMEC by the WT and mutant strains was evaluated (Figure 7B).
For the ΔlhrA and Δhfq mutant strains, we observed two bands corresponding to putative 5'-ends mapping to position −38 and –34 relative to the translation start site for lmo0302.
In contrast, while a reduced bacterial load of both mutant strains was observed in the kidney compared to the wild type this was not found to be significant (p = 0.075) (Fig. 4B).
Interestingly, in many mutant strains we observed substantial production of nuclear 5S rRNA and detected some minor amount of newly synthesized 5S rRNA in the cytoplasm (Fig. 5, lanes 8,10,12,14,26,28,30,32), leaving open the possibility that in yeast 5S rRNA containing particles can translocate to the cytoplasm independently of most of the residual LSU components (see also discussion in [51]).
In both of the ptrn-1 mutant strains, we observed fewer total EBP-2 GFP EBP-2 GFPthan in the wild-type strain, but the direction of EBP-2::GFP movements was like than of wind-thee.
As previously shown, wildtype U112 acid phosphatase activity is predominantly cell associated with little activity in the culture supernatant; whereas, acid phosphatase activity of the fopC mutant strain is observed to be predominantly in the supernatant, with little cell associated activity (Fig. 7B).
However, the reduced mean lifespan seen in the mutant strains was not observed with RNAi of dhhc-1 and - 14. Combined knockdown of dhhc-2 with - 8 and ppt-1 with ath-1 gave a reduction in mean survival time.
However, obvious differences in hyphal tip morphology or internal organelle structure in the hyphae of the four mutant strains were not observed using the plasma membrane endocytic dye FM4-64 or an ER dye (ER-Tracker Red) under either VMM or Avicel conditions (Additional file 2: Figure S1C and D).
