To determine whether the H4G94P mutant had normal cell cycle progression, we examined the DNA content of an asynchronous population of yeast cells by flow cytometry.
However, in vivo growth assays in both murine and rabbit CNS showed that the mutant and wild type Cryptococcus had similar growth rates, suggesting that the mutant had normal growth in the brain [ 9].
Consistent with normal myelination, the mutant had normal g ratios at 3 weeks (0.61 ± 0.02 and 0.64 ± 0.01, mean values ± SEM, n = 3 for control and mutant respectively, p = 0.24) and normal axon diameters (2.65 ± 0.05 μm and 2.55 ± 0.09 μm, n = 3 for control and mutant respectively, p = 0.43).

Mpk1 is increasingly phosphorylated with elevated temperatures and the levels are similar between the wild-type and cdc48-1 cells (Fig. S5A), showing that the cold-sensitive cdc48-1 mutant has normal response to heat shock.
As previously reported, starch granules of the Atss3-1 mutant have normal morphology [ 29].
The mutant has normal kinase activity and interacts with GRB2, but can no longer phosphorylate XPB.
All mutants had normal number of DD neurons in L1 larvae (data not shown).
The pal1 and pal2 mutants had normal lignin levels, presumably because of gene redundancy.
By contrast, nhsl1b mutants had normal neuroepithelial and floor plate planar polarity (Fig. 5D,F-H).
However, other spl mutants had normal cauline stem lengths (data not shown), while spl15 mutants had slightly stunted stems (Additional file 3) as reported previously [ 13].
