Our results unequivocally demonstrate that LRRK2 transgenic mice develop motor impairment consisting of small anomalies in rotarod performance (presumably reflecting a deficit in motor coordination and dystonia) and a strong deficiency in the hanging-wire test (reflecting muscle weakness), rather than hypokinesia which was difficult to be demonstrated in the actimeter.
Post-training maximal inspiratory pressure was independently associated with weaning success, suggesting that respiratory muscle weakness rather than fatigue is a greater contributor to weaning failure.
Greater difficulty (high values of Yinf, the final instability rate) to maintain erect posture appears to be mainly associated with plantar-flexor muscle weakness, rather than to damage of the proprioceptive system.
The correlation between Yinf and leg strength score (a global measure of muscle weakness) seems to indicate that the difficulty of the CMT1A group in maintaining quiet standing is associated with muscle weakness, rather than with the proprioceptive loss of large sensory nerve fibres whose index (vibration sense) did not correlate with Yinf.
The impact that a tear can have upon muscle strength, in terms of muscle weakness rather than pain inhibition, was mentioned by some, but not the majority, of participants; "it's mainly the weakness" (I/V 18), "I can't lift any weight" (I/V 9).
This is consistent with a recent analysis of muscle biopsies from NEB-related nemaline myopathy patients that showed myofibrillar dissociation appeared to be an important cause of muscle weakness rather than the frequency or position of nemaline bodies within muscle fibers, which inversely correlated with severity [ 28].

These results indicate that the rotarod performance deficit of D/+ mice does not result from muscle weakness, but rather from impaired coordination.
Until now many prognostic studies including people with ICU-acquired muscle weakness used rather a traditional prognostic design using a baseline test and compared with ICU discharge and follow-ups 5 28 29 and only some studies measures continuously over time.
These observations are in principle similar to those our own group and others have made when using volitional measurements of respiratory muscle strength whereby a high value supports confirmation of preserved muscle strength and a low value is not necessarily representative of muscle weakness, but rather is related to ability to perform the test effectively [ 26- 29].
Fatigue has a similar detrimental effect on all patient groups (P = 0.59) except for patients with hyperkalemic periodic paralysis in whom muscle weakness and myotonia rather than fatigue most affect their QoL perception.
To the best of our knowledge, many prognostic studies including people with ICU-acquired muscle weakness used a rather conventional prognostic design using a baseline test and compared with ICU discharge and follow-up data 6 36 37 and just some studies measured functional recovery continuously over time.
