Haldane favored the single mutation mechanism, but Fisher and Wright backed multiple gene change.

Lung cancer is a major cause of death throughout the world and develops slowly over many years from sequential accumulation of multiple gene changes in pulmonary cells (Zochbauer-Muller et al, 2002).
For our study, we compared only a handful of genes that would generate only three false positives, and we found alterations in most of these genes; for interpretation, we relied on multiple gene changes in a given pathway, as well as effects that repeated across different treatments and/or different time points, rather than changes in any one gene.
Genes encoding enzymes with the same function (isozymes) can also be problematic: often the quantity of real interest for a particular reaction is the net change in flux or flux capacity for that reaction, thus multiple gene changes related to a single reaction should be accounted for together rather than as separate nodes in a graph representation of metabolism.
This predictability of individual gene expression on a global background of multiple gene expression changes argues for a predominantly non-random, rather than stochastic, nature of reprogramming errors in bovine SCNT embryos which is in agreement with the study of Inoue et al. [ 39] on mouse SCNT.
Since conserved gene-level changes are rather rare events, differ between individuals, and methodologically difficult to assess, pathway-level analysis detects multiple relevant gene changes in different individuals within the same phenotype (here, PD), which consists of the entire repertoire of all responsible changes eventually leading to the same phenotype: PD.
Down-regulation of Nanog and Sox2 gene expression can change multiple gene expression patterns and signal transduction pathways, and will initiate ES cell differentiation.
The central hypothesis guiding this research is that normal biological and physiological pathways and gene expression signatures are varied in resistant insects through changes in multiple gene expression, thus enabling resistant house flies to adapt to environmental or insecticidal stress, and that these changes are controlled by a regulatory network and perhaps by signaling transduction.
For instance, offspring of rats fed a restricted protein diet throughout pregnancy showed changes in DNA methylation at multiple genes, with corresponding changes in gene expression, both of which were prevented by maternal supplementation of folic acid (an important methyl donor), further implicating an epigenetic mechanism [22], [23], [24].
To ensure that the absence of a marker gene expression faithfully reflects a swimbladder defect rather than simply a down-regulation of its expression, it is desirable to use multiple gene markers to trace tissue changes.
To date molecular profiles may consist of multiple gene mutational analyses, gene copy number changes by fluorescence in situ hybridization (FISH), gene expression profiles measured by microarrays (MA) and protein expression by immunohistochemistry (IHC).
