To investigate whether it was possible to reverse the aberrant pattern for LPL reactivity in islets of ob/ob mice towards the pattern seen in WT mice, ob/ob mice were treated with daily injections of leptin for 12 days.
Indeed, immunfluorescent staining of skin samples from HDGFTyr/HDGF-/ in comparison to HDGF-/ mice proofed that the distinct staining pattern seen in HDGFTyr mice originates from the transcript of the Tyr-HDGFgene construct.
Of the tissues investigated in that study, including skin, liver, muscle, and testes or ovary, the muscle was the most lethal to mice, similar to the pattern seen today.
This vascular PrPres distribution in tg44 mice differed markedly from the pattern seen in non-transgenic C57 mice expressing anchored PrP, where PrPres was not in a perivascular location, but instead tended to cluster around parenchymal cells with large nuclei, possibly neurons or astroglia, as has been reported previously [ 34].
Nevertheless, the pattern seen in 03A-0241; Tg(tetL)1Bjd/J mise is unique to these mice, as it is not seen in carriers of other T2/GT2/tTA or T2/GT3/tTA insertions (with or without associated concatemers) when crossed to the Tg tetL 1Bjd/J strain (above and data not shown).
In contrast, the leftward shift in platelet velocity pattern seen in WT mice after treatment with TNFα was much more prominent in TNFR1−/− mice, indicating more interaction of platelets with the endothelium.
However, the percentage of CD4+CD25+Foxp3+ T cells increased significantly after the infection (Fig. 6B), following the same pattern seen in suckling mice (Fig. 3A).
This pattern of PrPres distribution did not appear to follow the blood vessels and was markedly different from the perivascular plaque pattern seen in tg44 mice.
This is analogous to the distinction between the conservation of global network topological properties and the divergence of local connections previously observed for orthologous human and mouse coexpression networks [ 6], but the pattern seen here is even more extreme.
