Among the most significant replicating models was rs12597188 (intron of CDH1)–rs11564445 (intron of CTNNB1).
Figure 2 shows the P-value and hazard ratio (HR) distribution (Cox proportional hazards model) of all drugs after selecting the most significant replicate.
The intronic SNPs of EGF and EGFR, rs3796947 and rs6954351, respectively, were among the five most significant replicating models in the discovery dataset.
Our most significant findings were replicated in silico using two independent data sets, with the combined meta-analysis revealing markers below the threshold for genome-wide significance, namely, P= 7.8 × 10−12 for rs964184, P= 1.4 × 10−10 for rs2108622 and P= 8.2 × 10−9 for rs11057830.
However, GWAS always focused on the most significant variations when replicating the associations, and reported genes according to physical locations of the genome.
Particularly, the most significant SNP, rs10955255, was replicated in two of nine independent sample sets; and the odds ratios (ORs) of all nine sample sets had similar implications.
Of note, we also replicated the most significant SNP (rs11891311) reported in East Asians and Europeans.
We further replicated the most significant associations of SNPs with SLE using 2,612 cases and 2,323 controls of Asian ancestry.
Regarding the LFEC-related results for OAR6 that were obtained by LA, LDLA and LDA, in the current study, we replicated the most significant QTL that was previously identified through a microsatellite-based genome scan using a different set of Churra sheep half-sib families [ 20].
