In histologic and morphometric analyses, the distribution of particles, the degree of inflammation, epidermal thickness, and the inflammatory infiltrate were characterized and quantified by standard and special histological stains as well as immunohistochemistry for CD3+ lymphocytes.
For morphometric analyses, the values found in control and specific siRNA-transfected cells were analyzed by using paired t-test.
The new, well-preserved, late Miocene fossil B. wenshanensis is hypothetically dated at ca.11.6 Ma according to the International Stratigraphic Chart 2010 (www.stratigraphy.org), so we used this new fossil to calibrate the crown age for the results of the morphometric analyses and the phylogenetic relationships on the basis of the ITS set (Additional file 3: Table S3).

Based on our results, morphometric analyses support the genetic distinction of the two populations and also reveal that the level of genetic variation is associated with morphological shell shape variation in both populations.
