In Dicer1-hypomorphic mice, the size of the pancreas in adults (8 10 weeks of age) was nearly identical to that in the wild-type mice; however, there were more small islets (Fig. 4).
Fewer large islets (s>250 µm) and more small islets (100∼250 µm) than the expected number from the lognormal distribution are explained by the occurrence of islet fission, which is also supported by the morphology of large elongated islets in Fig. 1D.
Thus the duodenal lobe contains more small islets than the gastric lobe.
In pregnancy there were more small islets rather than an increase in islet size or beta cells per islet.
From the same group, Minh et al. (abstract 4) obtained pancreata at autopsy from six nondiabetic pregnant women and nine nonpregnant women, matched for age and prepregnancy BMI, showing a tripling of β-cell mass from week 20 to 40 of gestation with significantly more small islets and increased numbers of cells positive for insulin in pancreatic ducts without change in β-cell size.

The post-partum women retained the pattern of having more (but small) islets than did non-pregnant women, implying (1) that these putative new islets are not immediately and selectively lost after birth; or (2) that they formed partly in response to obesity.
Each sample contained more than 450 small islets or 50 large islets from 1 rat When the blot densitometry was normalized by IE, Glut2 levels were significantly higher (p < 0.05) in small islets than in large islets (Fig. 5c).
Normalization by IE indicates that there is statistically more DNA and total protein/IE in small islets compared to large (Tables 2 and 3).
The inverse correlation between total cell number per IE and islet size can explain this finding, because when the total cell number was normalized to IE, there were fewer cells per IE in large islets compared to small islets, which might be due to more intracellular space and vessels in the large islets as described previously [ 31].
However, when normalized by cell number, large and small islets had no difference in Glut2 levels, but large islets contained more proinsulin.
However, using the IE-based volume calculation, more IE of large islets was needed to isolate the same amount of protein compared to small islets (p < 0.05).
