The long sequence reads provided by Third Generation Sequencing techniques such as the Pacific Biosciences SMRT technology should allow more reliable assemblies of PKS and NRPS gene clusters and, in principle, yield single contigs for all of the replicons present in an actinomycete.
The better the quality of the trimmed sequences, the more reliable the final assembly.
