Comparing Fig. 4a, d shows the case when there are more clusters with a smaller variance (larger (alpha)).
In order to improve the amplitude SNR by a factor of two or more, clusters with at least four members were used in estimating the location of AE.
This results in producing more clusters with some of them having centers close to one another.
We reasoned that algorithms designed to map the coordinates of template clusters may fail to distinguish two or more clusters with the same barcode when they are in very close proximity (Fig. 1b).
