Quantitative analysis showed that most PSA-NCAM+ cells (61%; 360+/−38,9 cells/10 mm see Figure 1R and Table 1) reside in the molecular layer, the remaining percentage being widespread in the granule cell layer (35%; 200+/−42,1 cell/10 mm) and, to a lesser extent, in the white matter (4%; 20+/−2,9 cells/10 mm) (Figure 1R).
Underlying sedimentary layers remain but lack the same profile.
They showed a piriform cell body (15 20 µm long), a long, thin axonal-like process with a U-shaped direction remaining within the molecular layer, and one to three thicker, dendritic-like processes on the opposite side of the cell (Figure 1G,H).
In contrast, the ATXN1[ 82Q] -FVB;SV-129;C57BL/6 mice had a slightly thinner molecular layer that decreased rapidly by 12 weeks of age and remained at 115µ m from 16 to 20 weeks of age (Fig. 2B).
However, the remaining three neurons had somata and dendrites located in the molecular layer and were therefore termed molecular layer cells (MLs; green code in Figs. 2A and 6; Supporting Information Fig. 6), corresponding to MOPP cells (Houser und Escalpez, 1994; Ceranik et al., 1997) and neurogliaform cells (Armstrong et al., 2011).
The population, dendritic territory, and axonal processes of Purkinje cells remained normal in the cerebellum of treated twitcher mice, where radial fibers of Bergmann glia spanned the molecular layer and collateral branches ensheathed the dendritic processes of Purkinje cells.
