The mode of action of such integrin-mediated "survival signaling" known so far is largely through the modulation of signaling and/or transcriptional events that counteract death-inducing pathways16,57,58.
In particular, rocaglates are being investigated as potential therapeutics for a variety of cancer types including leukemias, melanomas, myelomas, lung cancer, breast cancer, colon cancer, prostate cancer and glioblastoma due to their multiple mechanisms of action; induction of apoptosis via protein synthesis inhibition, modulation of signaling pathways, cell cycle regulation, etc.21,23.
More broadly, this work suggests that bulky ligands could be used to drive the accumulation of specific receptors at the plasma membrane surface, providing a biophysical tool for targeted modulation of signaling and metabolism from outside the cell.
