In cultured cells, Sherpa mediated ubiquitylation of dMyd88 and Sherpa itself, and Sherpa and Drosophila SUMO (small ubiquitin-like modifier) were required for the proper membrane localization of an adaptor complex containing dMyd88.
Interestingly, disease penetrance in the family with the corresponding mutation [ 6], as well as in most other described DCM families, is incomplete, indicating that a "second hit" (environmental or a second genetic modifier) is required for disease development and progression.
Therefore, the aforementioned antimotifs that each form a circuit of a quick originating regulator aided by a slower modifier (i.e. intermediary regulator) seem to be inappropriate for fine-tuning of the target gene, possibly because the modifier is required to monitor the originating regulator at a higher frequency than the switching frequency of the originating regulator.
The absence of binding to the pure PT surface shows that electrostatic attraction to the modifier is required to promote adsorption, which will limit the positively charged porphyrin to adsorbing only to sections of the surface where it overlays at least one of the negatively charged MPS modifiers.
These benign tumors can then be used to identify genetic modifiers that are required for either initial tumor development and/or metastasis (Fig. 2A).
It has also been suggested that mutations in modifier genes are required for full penetrance of Bardet-Biedl Syndrome [ 62- 65], making it possible that the observed bbs7 MO phenotypes may be specific to the genetic background in which they were observed.

Consistent with this, many H3K4 methylation modifiers and their components are required for ES cell self-renewal or differentiation.
We propose that chromatin modifiers, such as HDACs, are required for establishing a DC gene network, where Flt3/STAT3 signaling drives PU.1 and IRF8 expression and DC development.
