Numerous studies have reported that modification of cholesterol content can affect APP processing.
In particular, it is not clear whether the modification of cholesterol homeostasis in AD brains is a cause or a consequence of the disease.
To investigate whether this control of neuronal activity by APP was related to modification of cholesterol turnover, calcium oscillations were analysed in neurons treated by mevastatin, an inhibitor of HMGCR.
Of note, whereas abnormalities in cholesterol metabolism are tied to a derangement of cholesterol synthesis and uptake in the peripheral tissues, leading to increased 'total' plasma cholesterol, that is, hypercholesterolemia, in many cases, they also appear to involve oxidative modification of cholesterol and/or altered cholesterol homeostasis within the brain.
Thus, although some experimental evidence indicates that alterations in cholesterol metabolism in the brain might contribute to the pathogenesis of AD (Kojro et al., 2001; Simons et al., 1998), it is not clear whether the modification of cholesterol homeostasis in AD brains is a cause or consequence of the disease.
As an example, enzymes CYP11A1 and CYP17 from the CYP450 family participate in the modification of cholesterol to yield T, 62 which is then converted to biologically more active DHT by steroid 5α-reductase type 2 enzyme (SRD5A2) in the prostate.
Each cholesterol category was considered to be managed if documentation of any of the following was found: (a) the need to recheck an abnormal cholesterol level; (b) discussion of lifestyle modifications; or (c) prescription or adjustment of cholesterol-lowering agents.
We suggest that modification of cellular cholesterol content with Dynasore protects cells against the effects of CDCs.
