Immunohistochemical staining of each sample by 1G8s was assigned a score of 0 to 3+ as follows: 0, no stain to less than 30% of cells staining faintly; 1+, greater than 30% of cells staining light to moderate; 2+, greater than 50% of cells staining moderately; 3+, intense staining of majority of the epithelial population.
L.M.D. and R.M.N. scored each core by visual microscopic inspection as follows: 0+ for no staining and no background; 1+ for weak blush of cytoplasmic staining; 2+ for moderately intense granular cytoplasmic staining; 3+ for markedly intense granular cytoplasmic staining.
Quantification was made as follows: <33 % of cancer cells 1; ≥33 to 66%% of cancer cells 2; >66 % of cancer cells 3; absent/weak staining 1; moderately intense staining 2; strong staining 3.
Thus the immunostaining results were divided into 4 categories: negative (-; lack of specific staining); mild staining ; moderate staining ; intense staining.
The slides were evaluated as no staining; 1+ (cytoplasmic staining or discontinuous membrane staining); 2+ (membrane staining with moderate intensity), and 3+ (intense staining with retained membranous staining).
The following scoring system was used: 1+, weak staining; 2+, moderate staining; 3+, intense staining.
Moderate staining in almost all carcinoma cells was detected in the highly expressing HCC), whereas the moderately expressing tumour exhibited very intense staining in a restricted number of carcinoma cells).
SOR amnion epithelium exhibited moderately intense and extensive MMP-9 staining that was significantly greater compared with the SCS.
Staining of lung tissues also indicated the presence of MICA positive cells; however, compared to the tracheal epithelial cells this staining was only moderately intense (Figure 5B).
