L.M.D. and R.M.N. scored each core by visual microscopic inspection as follows: 0+ for no staining and no background; 1+ for weak blush of cytoplasmic staining; 2+ for moderately intense granular cytoplasmic staining; 3+ for markedly intense granular cytoplasmic staining.
Ultrastructurally, the inclusions were found to be moderately electron-dense, finely granular, homogenous bodies that are encircled by membranes, corresponding to a dilated endoplasmic reticulum.
Nfatc3 mRNA was moderately expressed only in the granular layer of the dentate gyrus and was detected at low levels in the CA1 CA3 pyramidal layers (Figs. 4E and F).
Nfatc1 and Nfatc4 were moderately expressed only in the granular neurons (Figs. 4A, 4G, 5B, and 5K), whereas Nfatc2 and Nfatc3 were expressed in the granular cell layer and also in the Purkinje cell layer (Figs. 4C, 4E, 5E, and 5H).
An IHC score was assigned to each case according to the following criteria showing the designated staining pattern: 3+, intense, granular cytoplasmic staining; 2+, moderately, smooth cytoplasmic staining; 1+, faint cytoplasmic staining; and no staining.
Pileus 35 60 mm in diameter, pinkish brown-to-brown, moderately smooth, sprinkled more or less evenly (slightly more dense at centre) with tiny, dark, granular scales, convex at first, becoming plano-convex, dry, commonly with a broad, sharply demarcated hygrophanous (but not viscid) zone around the cap perimeter, not striate or with faint striae only at the margin; flesh off-white to pale pink.
Although in the epidermis there were no gross phenotypic abnormalities or barrier dysfunction apparent at birth, histological examination revealed that the newborn (Figure 2A) and 1 week-old (not shown) transgenic epidermis was moderately thicker than that of the wild type, with a thicker stratum corneum, a less-compacted granular layer and an overall expanded suprabasal compartment.
In the cerebellum P12, ANP32A was expressed moderately in only the nucleus of the Purkinje cells, which scattered evenly between the molecular layer and the granular layer.
