The step index profile gives rise to a single BSSW mode while the gradient index profile has three BSSW modes due to the varying band edge within the multilayer.
Among the tested first order tetrahedra, the nodally integrated tetrahedron with an additional bubble mode in the gradients was found to be most accurate.
The first-dimension reversed-phase separation was performed on two serially connected 150 × 2.1mm Inertsil 5 ODS3 columns, particle size 5μm, operating in the gradient mode (Varian, Middelburg, The Netherlands).

In the design of the gradient mode, we chose a mobile phase composition of 75% methanol in the first 5 minutes of the run to allow sufficient peak resolution of the amide and the ester peaks, and subsequently a gradient with increasing methanol content up to 95% was set over 10 minutes followed by holding for another ten minutes (Table 1).
Separation was achieved on Acquity BEH-shield RP18 UPLC column (3.0 mm × 100 mm, 1.7 μm) under the gradient mode of elution by using mobile phase A (0.02M diammonium hydrogen phosphate/acetonitrile, 80 20 v/v) and mobile phase B (0.02M diammonium hydrogen phosphate/acetonitrile, 30:70 v/v).
The gradient mode proved to be stable according to method validation results.
In addition, we considered the appearance number of modes, as well as the gradient magnitude in order to optimize the performance of intra mode detection.
