Techniques such as atomic force microscopy (AFM) can be used to generate extremely high-resolution images of ECM structure (Graham et al., 2010) and, with the upcoming force-mapping modality experiments that many atomic force microscopes are capable of, local force measurements can be taken to evaluate matrix elasticity and stiffness at the micron scale.
