This PCR was performed in a final volume of 100 μl, containing 0.25 μl of library, 50 mM KCl, 10 mM Tris-HCl, (pH 8.8), 1.5 mM MgCl2, 0.1% Triton X-100, 0.2 mM concentration of each dNTP, 0.5 unit of Pwo DNA polymerase, and 100 pmol of each primer.
The antibody gene libraries HAL9/10 were packaged separately inoculating 400 mL 2 × YT-GA (2 × YT containing 100 mg/mL ampicillin and 100 mM glucose) with a library glycerol stock.
Screening methods were as follows: first, low-molecular-weight compounds (2 mM DMSO stock solution) from library plates (the final concentrations of the compounds were 0.25, 1, 3, 10, 30 μM) were diluted with HSip-1 and 3MST solution to 20 μM solution (10% DMSO) with a Multi-Drop Combi.
Cells were incubated overnight, followed by 30 min pretreatment with 10 µM compounds in DMSO (10 mM stock concentration from The Spectrum Collection library).
Wells were filled in order with 45 μl of assay medium, 0.2 μl of library compounds (10 mM in DMSO) and 45 μl of assay medium inoculated with 1×10 or 1×10 cells/well for the –Ade or +Ade assays, respectively, of the assay strain grown without Aβ-MRF overexpression.
We amplified a cDNA library from six samples of 0 mM treated plants and six 300 mM treated samples.
Most compounds from this library are provided at 10 mM, however some peptides from the library are supplied at a concentrations of 1mM.

The 5' U preference was maintained in individual mycelial libraries – CM, MM, NS and PQ, but the CS library contained more sRNAs starting with G, closely followed by U. The suppression of C at the 5' position was also maintained in most of the mycelial libraries except for MM and PQ (data not shown).
The number of 150 mm plates was calculated based on the library titer.
Only compounds totally soluble at 10 mM in DMSO were included in the library.
