Despite the recent progress in the field of reconstruction and regularization techniques, the missing wedge artefacts remain untreated in most current reconstruction workflows in structural biology.

Consequent artefacts, such as blurring and elongation of features in the missing wedge direction, deteriorate the quality of the reconstruction and make quantification difficult (e.g. [12]).

This amplitude decrease with N should not be interpreted as a real effect: artifacts from the missing wedge induce a deceptive contrast into the reconstructions, with for instance white fringes around the darker features in the XY view (Fig. 4A, FBP N = 1).

Pure image-processing techniques such as NAD filtering or machine learning classification will not be able to compensate for missing wedge elongation this is an issue which would be more effectively solved by the reconstruction algorithm.

For a reliable assessment of the method, tomograms must be generated by simulating the actual tomographic image reconstruction process, allowing the inclusion of noise, tomographic distortions due to missing wedge and electron optical factors such as contrast transfer function (CTF) and Modulation Transfer Function (MTF) (Fig. 4).

Hessian, Gabor and structure filters detect the orientation of image features one very specific usage case for these is when compensating for missing wedge artefacts.

Due to the missing wedge artifact, thylakoid grana that may have been present at the top and bottom of the chloroplast could not be resolved.

Less likely but still possibly, connections might have existed but were not observed due to the missing wedge artifact or limits in resolution.
