Despite the recent progress in the field of reconstruction and regularization techniques, the missing wedge artefacts remain untreated in most current reconstruction workflows in structural biology.

Hessian, Gabor and structure filters detect the orientation of image features one very specific usage case for these is when compensating for missing wedge artefacts.

Due to the missing wedge artifact, thylakoid grana that may have been present at the top and bottom of the chloroplast could not be resolved.

Less likely but still possibly, connections might have existed but were not observed due to the missing wedge artifact or limits in resolution.
