Scanning electron microscopy of spheres obtained from Hbt. salinarum NRC-1, which were fixed with glutaraldehyde after dissolution of halite crystals (see Methods), showed mainly single globular particles (Fig.B,D,F) with a diameter of 0.40 ± 0.02 (SD) μm.
Time-lapse fluorescence microscopy of whole spheres plated onto a laminin substrate in the absence of growth factors revealed the same three morphologically-distinct cell types as those observed in dissociated cultures.
The resultant particles show four distinct morphologies of sphere, hollow, flake and porous flake using scanning electron microscopy, transmission electron microscopy and nitrogen adsorption and desorption isotherms.
Scanning electron microscopy results show that small cavity exists in the center of sphere.
An estimation of sphere size was made in vivo by passing sphere suspensions of Hbt. salinarum NRC-1 (ca. 10 CFU mL−1) through filters of pore sizes 0.2, 0.45, 0.8, 1.2, and 5 μm (Minisart; VWR, Vienna, Austria), respectively, and examination of recovered cells in the filtrate by subsequent microscopy, using phase contrast.
Quantification of sphere growth and passaging.
For determination of sphere size, we gauged sphere diameter optically with an object micrometer.
Negative-stain electron microscopy of purified SFTSV virions shows that the virus particles are enveloped spheres or ovals with diameters of 80 100 nm.
