An awareness of TNF began with its detection in the serum of mice receiving Gram-negative bacterial endotoxin, that is, LPS, several weeks after they were infected with Bacillus Calmette Guérin (BCG), an attenuated strain of Mycobacterium bovis.
One interesting point is that the median of the absolute number of mutants actually remains constant if we compare the Fos-R mutant load in mice receiving fosfomycin or not, although the total bacterial count decreased by two orders of magnitude in the dosed mice.
Alternatively, for mice receiving a dose of 1 × 10 CFU, the original bacterial stock was diluted in PBS to 1 × 10 CFU/mL, and 100 μL was injected per mouse as above.
This bacterial stock was directly used for systemic inoculation of mice receiving a dose of 1 × 10 CFU by injection of 100 μL into the lateral tail vein using a 0.5 mL syringe with a 29G needle (Becton Dickinson).
Accordingly, peritoneal leukocytes of fibrate-treated mice contained significantly more S. typhimurium (Fig 2), whereas conversely, bacterial counts in peritoneal exudates of fenofibrate-treated mice were dramatically lower than in mice receiving a control diet (Fig 2C).
Mice that had been vaccinated with MOMP in CAF01 were partially protected and exhibited decreased bacterial load at all time points post infection compared to both un-vaccinated control animals and mice receiving MOMP delivered in Al(OH 3 (Fig. 6E).
In order to assess the duration of the CAF01-induced protection, bacterial growth was monitored at different time points (week 4, 6, 12 and 24 after infection) after infection showing the maintenance of approximately one log reduction in bacterial numbers throughout the course of infection at a level almost comparable to that seen in mice receiving the live BCG vaccine (Fig. 3G).
Similarly as observed in C57BL/6 mice, Pparα−/− mice displayed significantly enhanced neutrophil influx to the site of infection 12 h after bacterial inoculation (Fig 3A) and reduced bacterial loads in liver and spleen after 3 days of infection when fed with fenofibrate as compared to mice receiving a control diet (Fig 3B and C).
Furthermore, septic mice receiving normal saline experienced significantly more post-sinusoidal adhesion than septic mice receiving any other crystalloid fluid.
Methods: C57BL/6 mice were infected with Helicobacter felis and received bacterial eradication therapy after 2, 6, or 12 months of infection.
