The fluorescent molecule 5 detects Aβ plaques in brain hippocampus sections of transgenic mice, while also visualizing CAA.
For images of the full hippocampus, sections were stained with antibodies against PROX1 (rabbit polyclonal #ab101851, Abcam, 1 500), CTIP2 (as above), and NEUN (mouse monoclonal conjugated to AF488 #MAB377X, EMD Millipore, 1 200).
To accomplish this, we incubated coronal sections of mouse hippocampus with a rabbit monoclonal anti-BACE1 antibody and goat anti-rabbit IgG conjugated to ultrasmall gold particles followed by silver enhancement, as previously described [ 60, 111].
Sections of each mouse hippocampus were imaged together and the number of astrocytes per mm, average astrocyte area per μm, and relative intensity of GFAP staining were measured by the Image J software (NIH).
For densitometric measurements and quantification of pixel densities of anti-PTK2B, anti-PTK2BpY402, and anti-MAP2A immunolabelled sites in neuronal dendrites in the stratum radiatum of the hippocampus CA1 region, measurements of pixel densities were obtained from within three equal-sized areas (3128 μm) from three sections from each mouse hippocampus using the Photoshop program.
