Compared with classical molecular biology protocols and cytogenetic analyses used as reference standards, results of the 26-plex RT-PCR method were concordant in all 204 (100%) cases of adult AML tested.
This high level of concordance mirrors that observed in our previous analysis of X-linked gene expression in TSCs, where allelic biases determined via RNA-Seq and an alternate method were concordant in 18 of 18 assays (nine genes in two TSC lines) (Calabrese et al. 2012).
The objectives of MTMM analysis are to determine whether the measurements of each trait derived by multiple methods are concordant, to show that measurements of different traits obtained using the same method are discordant, and to estimate the influence of different methods on the measurement of traits.
On the other hand, when PSB was performed without antibiotics, direct examination was positive in 65% (n = 46), culture was positive in 62% (n = 43) and those methods were concordant, non concordant and partially concordant in 71% (n = 50), 18% (n = 12) and 11% (n = 8), respectively.
When PSB was performed under antibiotic treatment, direct examination was positive in 26% (n = 12), culture was positive in 28% (n = 13) and those methods were concordant, non concordant and partially concordant in 93% (n = 43), 4% (n = 2) and 3% (n = 1), respectively.
The estimates obtained from both methods were concordant.
Results obtained from both methods were concordant in 183 (82.8%) cases (κ=0.48).
However, a significant nonzero correlation coefficient does not imply that two methods are concordant [ 23].
In 433 patients (86%), however, the two methods were concordant in weighting the "other risk factors and disease history" with a weighted κ value of 0.764.
In 9 out of the 12 responders (75%), the results of the LV d P/d tmax and PEAarea methods were concordant (Table 2 ).
