Both adherent and floating cells were collected and fixed in 100% methanol for at least 1 h at −20°C.
For immunofluorescence microscopy, the harvested cells were washed in PBS buffer and fixed in cold methanol for at least 20 minutes at −20°C.

For immunofluorescence, cells were fixed for 10 min either with 4% paraformaldehyde at room temperature or with a 1 : 1 mixture of acetone and methanol for at −20 °C (for γ-tubulin), and stained with mouse anti- α-tubulin (B-5-1-2, 1 : 2000), mouse anti- γ-tubulin (GTU-88, 1 : 1000; Sigma-Aldrich), and rabbit anti-phospho-histone H3 (06-570, 1 : 200; Millipore, Billerica, MA, USA).

Pretreatment steps were performed for some antibodies as follows: 2N HCl for 30 min at 25°C for BrdU and 100% methanol for 7 min at −20°C for MAG.
