Therefore, in this study we aimed to experimentally examine whether HNF4alpha take part in the metastatic tumor formation of HCC and its relationship with the MET markers.
Here, we investigated whether the reduced cell invasiveness following NAT1 knockdown in MDA-MB-231 cells was associated with mesenchymal epithelial transition (MET) by examining several well-characterized MET markers (Fig. 3A).
What we found suggested that HNF4alpha might play an important role in the metastatic tumor formation of HCC, and it might be related with the expression change of MET markers, or even the MET in the metastases.
Furthermore, based on Oncomine [ 10] data, we found that the gene expression analysis of the OVOL-TFs significantly correlates with the expression of MET markers in multiple cancer types.
We also confirmed the MET phenotype in these models by showing differential expression of critical MET markers including: up-regulation of E-Cad expression plus down-regulation of both vimentin and the EMT-inducing TF ZEB15 [ 3].
Statistical results showed that the expression of HNF4alpha was positively related with that of E-cadherin, and negatively correlated with that of Snail, Slug and Fibronectin, suggesting that the expression change of the MET markers in the metastatic lesions might be associated with HNF4alpha.
