Strikingly, we found that three specific human Rabs (endoplasmic reticulum/Golgi Rab2a, early endosomal Rab5a, and late endosomal/lysosomal Rab7a) strongly accelerated membrane aggregation of synthetic liposomes even in the absence of any additional components, such as classical tethers, tethering factors, and Rab effectors.

In this study we characterized the membrane aggregation capacity of penetratin using LUVs of three different compositions: PC and PE main phospholipids of, respectively, the outer and inner leaflets of eukaryotic cells and PG, a phospholipid highly represented in prokaryotes' outer leaflet.

On the contrary, the liquid disordered LUV, notably in the presence of PG, have shown membranes deformed by Penetratin interaction, confirming that the fluidity is required to observe the peptide induced deformations and high levels of membrane aggregation.

The Penetratin capacity to bridge membranes and produce LUV aggregation shows that in the presence of PG, membrane aggregation is clearly more efficient than in it is absence (Fig 3).

Relationships have been found between membrane aggregation and tube formation and size of membrane pores and the nature of the hydrophobic residues.

Membrane aggregation was stopped after 15 minutes of incubation.

How this affects the membrane-binding or membrane aggregation properties and thereby the function of ANXA10 is unclear.

Further reconstitution studies with guanine nucleotide-preloaded Rabs, the Rab-specific guanine nucleotide exchange factors, and the Rab GTPase-activating proteins will be required to more thoroughly assess the GTP/GDP-dependence of Rab-mediated membrane aggregation.

This indicates that the Rab-mediated membrane aggregation found here is a reversible process of membrane tethering and can be reversibly regulated by membrane attachment and detachment cycles of Rab proteins.

Moreover, we found that Rab5a and Rab7a had to be anchored on both, not either one, of two opposing liposomal membranes for driving membrane aggregation (Fig. 5B, lanes 3 and 7; Fig. 5C), suggesting that Rab-induced membrane aggregation is promoted by trans-Rab protein assemblies on apposed membranes.

The peptide-membrane interaction could be responsible for membrane aggregation by peptides bridging simultaneously two membranes or for negative curvature of the membrane asymmetry that can form tubes (Lamaziere et al. [2007]).
