EPCs were isolated from samples using Ficoll-Paque and five million cells were seeded on 6-well fibronectin-coated dishes (BD biosciences) in CFU-Hill medium (stem cell technologies, cat 05900) (Hill et al., 2003, Solomon et al., 2012).
There should be three medium sized stems in the middle.
The nucleated cells were grown in Mesencult medium (Stem cell Technologies, Vancouver, Canada) supplemented with 20% FBS, 1% penicillin-streptomycin and plated in 100 mm culture dishes.
Briefly, the nucleated cells were suspended at a concentration of 1×106/ µL in Mesencult medium (Stem cell Technologies, Vancouver, Canada) supplemented with 20% fetal bovine serum (FBS) (Hyclone, Logan, UT), 1% penicillin-streptomycin (Invitrogen, Carlsbad, CA) and plated in 100-mm culture dishes.
Then, PB-HP were committed to NK pathway expanding the cells in STEMα−Α medium (Stem Alpha, Saint Clement les places, France) supplemented with SCF and FL (both at 100 ng/ml), a condition that favours and enriches the frequency of NK progenitors [18].
The cell suspension was centrifuged for 10 min at 600 × g and the pellet mechanically dissociated 25 times in NeuroCult medium (Stem Cell Inc., Grenoble, France) using a glass Pasteur pipette and 20 times using 1 ml pipette tips.
Finally, dissociated cells were plated in standard neuro-sphere growth medium Neurocult proliferation medium (Stem cell Technology) supplemented with EGF (20 ng/ml) and FGF2 (10 ng/ml).
Pimmongkol et al. [ 10] stated that the width of vascular bundles and diameters of rice stems decreased in NaCl medium.
It was found that HA coating did not influence radiological results or clinical performance of these stems in the medium-term (Kim et al. 2003, Park et al. 2003).
For TiSiO2 the highest BMP-2 levels were recorded at 21 days of cultivation in OS medium, while in stem cell BMP-2 values increased slightly and in the OC medium the BMP-2 values decreased at 21 days.
