We have selected three probe sets with an average reading above 200 standard Affymetrix difference units [ 18] in the following conditions: (1) in all tissues, these are widely expressed genes; (2) in 20 to 29 out of 47 tissues (medium expression); and (3) in only 1 tissue (narrow expression).
We further divided the expressed gene into three categories based on their expression levels (low expression, medium expression and high expression), of which each category contains the same number of genes.
Color bar at the bottom represents log2 expression values, thereby green color representing lowest expression levels, black medium expression and red signifies highest expression level.
Color bar at the left represents log10 expression values; thereby green color representing lowest expression levels, black medium expression and red signifies highest expression level.
The scale for relative expression values is represented by color bars below the heat maps, thereby green color representing lowest expression levels, black medium expression and red denotes highest expression level.
Cytoplasmatic staining was classified as 1 (missing or weak expression), 2 (medium expression), and 3 (high expression).

Gene expression was measured by microarray analysis and then divided into four categories: no expression, low expression (log2‐scaled averages <4) and medium expression (≥4 and <8) and high expression (≥8).
We validated that microarrays have acceptable sensitivity and accuracy in detecting differential expression, especially for genes with high and medium expression levels and for detecting > 2-fold changes.
A statistical difference was observed in OS for the expression of TWIST1 (p<0.001): the five-year OS for each group was 79% 955% CI, 67% 91%) for those patients with low expression levels; 66.6% (95% CI, 51.9%–81.3%) for patients with medium expression levels; and 40.6% (95% CI, 24.6%–56.6%) for patients with high expression levels (Fig. 1B).
The genes with RPKMs ranging from 0 to 3 were considered at a low expression level; the genes with RPKMs ranging from 3 to 15 at a medium expression level; and the genes with RPKMs above15 at high expression level.
Those falling within one standard deviation of the mean were classified in the "medium" expression group, and those outside this range were deemed to have "high" or "low" expression.
